چکیده

Results TGF-β1 dose-dependently increased the contraction of collagen gel mediated by cardiac fibroblasts, reaching a maximal effect at 100 pmol/L TGF-β1. TGF-β1 also stimulated H-thymidine incorporation and total protein content in cardiac fibroblasts in the collagen gel lattice. TGF-β1 dose-dependently induced an increase in αsmooth muscle actin, a marker of myofibroblasts. The TGF-β1-induced reduction of area of the collagen gel was negatively correlated to the TGF-β1-evoked appearance of α-smooth muscle actin in the collagen gel matrix.

Transforming growth factor-beta 1-mediated collagen gel contraction by cardiac fibroblasts.

OBJECTIVE Myofibroblasts and transforming growth factor-beta(1) (TGF-beta(1)) are key elements of cardiac tissue fibrosis development. The aim of this study was to determine whether the ability of TGF-beta(1) to affect the contractile activity of cardiac fibroblasts depends on their differentiation into myofibroblasts. METHODS Cardiac fibroblasts (from male adult Wistar rats) from passage two...

The role of Smad3 in the fibrotic phenotype in human vocal fold fibroblasts.

OBJECTIVES/HYPOTHESIS To investigate the role of Smad3 as a regulator of transforming growth factor (TGF)-β1-mediated cell activities associated with fibrosis in normal human vocal fold fibroblasts. We also sought to confirm the temporal stability of Smad3 knockdown via small inhibitor ribonucleic acid (siRNA). Vocal fold fibroblasts were employed to determine the effects of Smad3 knockdown on ...

Matrix metalloproteinase-9 activates TGF-β and stimulates fibroblast contraction of collagen gels.

Matrix metalloproteinase-9 (MMP-9) is a matrix-degrading enzyme implicated in many biological processes, including inflammation. It is produced by many cells, including fibroblasts. When cultured in three-dimensional (3D) collagen gels, fibroblasts contract the surrounding matrix, a function that is thought to model the contraction that characterizes both normal wound repair and fibrosis. The c...

A 3D epithelial–mesenchymal co-culture model of human bronchial tissue recapitulates multiple features of airway tissue remodeling by TGF-β1 treatment

BACKGROUND The collagen gel contraction assay measures gel size to assess the contraction of cells embedded in collagen gel matrices. Using the assay with lung fibroblasts is useful in studying the lung tissue remodeling process in wound healing and disease development. However, the involvement of bronchial epithelial cells in this process should also be investigated. METHODS We applied a lay...

TGF-β and glutathione promote tissue repair in cigarette smoke induced injury

(CSE) on a three-dimensional (3-D) co-culture model with epithelial cells and mesenchymal cells to clarify how epithelial cells protect lung tissue from cigarette smoke in-vivo. Two types of gels were prepared. The one was the co-culture of human fetal lung fibroblasts (HFL-I) embedded in type-I collagen gel, with alveolar epithelial cells (A549) cultured covering the top of the gel. The other ...
